I believe the slot is what determines witch card is primary. You could try switching them, but I don’t think it will make any difference in performance.
If its working now play a while before you make any changes.

Just that the top slot with the x2 is the PCI2 x16 slot. The one beneath is the PCI2 x8 slot, having an x2 in the PCI 8.0 slot would decrease its performance surely due to bandwidth.

I’m not an expert on this but I think
Both cards will be running at X8.. Hopefully someone in the know will chime in on this…

I have a Bios Option which allows me to select my primary card.
Could you check yours? and the P45 only has x16 max So if you only use one slot it uses the x16 if you crossfire/SLI it divides to x8 + x8

Not sure that it matters as your programs will be running with all your GPU power.

This is going to sound very sad... but i've seen it happen. Make sure you have power to every single connector you can, at least 1 6 port per card. I spent an hour once trying to fix a machine before i actually looked at the machine's power cabling. I had asked the girl who built it if they were plugged in but on that day i learned human memory can fail... a lot.
