not just thermal compound/thermal pads... heatsinks will conduct electricity, so if it isn't seperated from the board by a layer of insulation, it will short the motherboard

But having a fan there and adding a heatsink to the back of the motherboard are two seperate things... adding airflow to a case is always good, but adding a heatsink to everything you can isn't
I assure you there is airflow coming inside the case through those holes at the back of the mobo

*sigh* alright, here's a simple explanation... air will not magically enter that vent and cool your motherboard... there is no fan to push the air... therefore, there is no airflow

But thats besides the point... stick a fan there, and its a much better way of cooling the back of your motherboard than to add a heatsink to the back...

All the exhaust fans inside the case create negative pressure , this pull air inside through every hole

There is ventilation even without a fan there
