I'm quite sure PCI will be fine for PhysX

Folding works fine on PCI-e 1x

PCI is fine for folding/Physx, not video rendering.

It is waste of money and performance by adding an extra video card. Because 9400GT is basically the same video card as 8600GT. These 4850 in crossfire will handle easily all the work. If you add an 9400GT, your performance will be worse than without it. Just let your 4850 do the work.

The 9400GT is inferior
It's more akin to the 8500GT

Btw, the 4850 doesn't support PhysX
