Over time your GPU could work its way out of the slot.

If it does that the best case is your notice strange behavior on you monitor and you have to reseat the card.

Worse case is the card gets so loose it falls out of the slot and that could cause some serious damage.

If I was you I would find a way to secure the card.

I'd like to add that when I was building my rig with the nVidia HAF X, I had problems screwing in my ASUS GTX 580 DirectCU II. The screws were very difficult to align correctly without pushing really hard. It has 3 slots as you know. If I wouldn't have been able to screw it in I would have RMAed the case. I think it needs to be screwed in for safety concerns.

I did get the middle in and screwed it. It was VERY HARD but I got the second screw in but I sort of lost the "click" of the GPU in the PCI slot. It was in the slot but the "click" when you release and push in the GPU was gone. I tried to put the third slot in, killed my fingers and no go. It just wouldn't do it. So I left it with just the two. I am more than confidant that it will hold up as I tightened them to hell. Of course, I will keep an eye on it.
