It is the PWM control signal wire. PWM is a more efficient way of controlling fans, but control via voltage will still work.



Black - ground
Red - 12v
Yellow - RPM
Blue - PWM

There's only one way to put the 4-pin connector into the 3-pin slot on a motherboard, so it'll be obvious which one is the "last one" when you plug it in.

Yes, you can. The plug is designed to work that way. The tab and slot will only fit in the right places. A 4 pin fan will work on a 3 pin socket, and a 3 pin fan will work on a 4 pin socket.

The last wire is not for reading rpm. In a 4-wire plug, you have

Black - ground (GND)
Red - 12 Volts (12v)
Color1 (often yellow, but varies) - RPM reporter (fan to motherboard)
Color2 (different from the other 3) - PWM control (5v signal from mb to fan)

The plugs were intentionally designed to be backwards compatible. Actually, they are also forward compatible: you can plug 2-wire, 3-wire and 4-wire plugs onto the same 3-prong and 4-prong headers. I have done all of these things many times.
