Or is it possible to remove it all together?

You can remove it altogether, not sure about moving it to a different spot.

It's as simple as unscrewing the back plate. However, moving it to a different slot may be possible if you're very precise about drilling holes for the removed hotswap back plate. That and it would only be possibe to place it in the top slot as there is nothing holding the drive sled beside the floor of the 5.25 bays.

Do you knwo the hotswap bay, is it literally just open it up and pop a HDD in? or do I need to wire it myself on the inside everytime?

To utilize the hotswap bay you need to attach a drive to a provided sled. It will then just slide into the backplate that has a standard SATA port and 4 pin molex wired internally. These only need to be connected when assembling the system.

Check my build log: http://www.overclock.net/t/1355177/build-log-project-shift-nzxt-switch-810/0_20

There's a part where someone asked for the full loop order from the res and back. However, it's most likely changed by now and by the time you finish reading the list, it'll most likely change again, lol.
The only thing I am not so sure of is using coolant vs. distilled water.

Read a lot about that, and you will never get the same answer from different people. Everyone’s loop is different and there are so many factors affecting it that what works for one person will not work for another.

I have found that the old standby of distilled plus silver kill coil is not the best, and some coolants are ok while others can cause issues depending on the specifics of your loop. I settled on distilled plus a good additive, I used Liquid Utopia.

I'd rather use Mayhems concentrates than using straight distilled w/kill coil. Distilled water alone does not prevent galvanic corrosion when mixing different blocks and rad metals. Also, when I removed my Swiftech CPU block, it didn't have any clogged channels or severely stain my XSPC tubing. Just my experience...
