D5 pumps - Do I need to use both the molex and the fan connector?

I have a new XSPC pump/res combo that came with two D5 pumps. The pumps have a 2 pin molex, but also a 3pin fan header with just one wire running into it. I would guess this would be to control the speed of the pump but it already has a manual speed setting on it. Do I need to plug in the 3pin connector or can I just use the molex only?

That the RPM line so you can tell the pump is running.

Yes. I have 2 D5 pumps in my "Muzzle Flash" build like that. I plug the DC power for both pumps to the power supply and the other connector on to a fan header on my motherboard, that way I can monitor the RPM of the pumps.
