As far as sealant goes, I just used an automotive high heat copper silicone and just put a small amount to cover the threads on any threaded barbs including ones with O-rings.

You probably don't necessarily need the high heat stuff considering the lower temps a WC loop has, but I had it already. No leaks here.

I use Permatex red RTV sealant just use it on the upper 2/3rd's of the barb..Clean it off with alcohol first to get rid of any oils on it..

Hey Trendy you get the Permatex RTV at auto parts stores...Its the red which is high temp...Just take off the O-Rings and put a good coat of it on about the top 2/3rd's of the barb it will have a little squish out around the top of the barb when you tighten it down I always just leave that like a little extra gasket...You have to be careful if you do that on anything made out of plexi because it could crack it out tightening it down that much...When you tube it all up if you have to twist the tubing to get it on always twist to the right so that you don't break the seal...

Thanks Ira, so I just put it on the threaded part of the barb and screw it in? Could I also apply some around the outside edge?

It will squish out around the outside of the barb I just leave it there to act like a gasket...Just put a goodly amount of it on the top 2/3rd's of the threaded part of the barb...And then tighten it down well just don't over-tighten say finger tight then another 1/2-1 turn....
