Orthos does not officially support quads, you will need to run two instances which can cause errors even on a stable OC.

Download: http://www.majorgeeks.com/Prime95_d4363.html

EDIT: There is no reason to run 4 instances of this. This version will automatically detect your cores and launch the threads needed.

Interesting I didnt know that about orthos. Thanks

I run two instances (loads four cores) with my quad and vista 64bit premium.

Its easiest to just run OCCT though as it will test up to 16cores at once.
I set the affinity to 1 on one instance and 6 on the other instance, could this be the problem?

How could you do it to 1 and 6, it should give you a option of four cores 1,2,3,4,, so really you could just set it to 1 and 2 then 3 and 4.

Still the easiest way is just use the linked P95 since it does all four or use occt.
