The P45 doesn't have PCI-E2.0. It only has 16 PCI-E lanes, which means you can run one card with full x16 lanes, or two with x8 each. x8 will bottleneck the card if you are rendering more than 2 million pixels(1920x1200). With one card, it won't bottleneck, with two, it depends on the resolution.

The P45 does offer PCIE 2.0.

Between the P5Q Pro and P5Q-E, aside from some BIOS options differences and the above mentioned difference in heatsinks on the board, the physical layout is quite different as well, which may or may not be the decision point for your particular setup.

they do have pci-e 2.0 lanes. When in CF those go to 8x wich equals PCI-E 16x. The performance loss is only minimal (about 5%)

Stay away from that board. It's garbage. Asus is poop too, for deceiving buyers. They put in a used board and called it "new". Ordered from newegg, got that board and it had a foxconn cover on the cpu slot. Like wt eff krnbbq? Asus is producing some rather trash boards right now, I'd stay away.

90% of boards are made by Foxconn

I've never had a problem with any of my Asus boards.

I've got the P5Q and so far it's been a great board, running now for just over 3 1/2 months with no problems. My e8400 overclocked easily to 3.6GHZ without having to change any setting in the BIOS except the FSB.

I had it and RMA'd it. No joke that 24 Pin placement will make you want to rip your hair out. I recommend the P5Q Pro or E.
