I believe it will, yes. But that's mainly because it'd slow the air down and you'd need more pressure to get air through the heatsink.

Oh and I was wondering how big these fans are? Sounds huge if it pushes that amount of air. And you'd need a separate PSU just for the fan

Really? Why not just add more fans. And about those massive 500rpm case fans, sure they push a lot of air, but it's not directed straight, it's dissipated to the sides. Take a massive heatsink and strap a high CFM fan to it, cooling will be awesome.

Besides, those fans used in automobiles aren't especially great around electronics.

You can exchange airspeed / air pressure

So you let the fan build up the potential energy and design the heatsink to ulitze it.

So essentially by making the least aerodynamic shape with the most contact surface area would result in a really good cooler.

hmmmm... this actually sounds like a bit of an interesting project. Just had a quick look at some of these; the OP has makes a good point.
12" thermatic vehicle rad fans looking at a range the average is ~$100 - 12v btwn 6&9amps, btwn 600-1000cfm.
I'm sure those big CFM ones would be obscenly loud, but nothing a potentiometer can't fix to wind down the current a bit.
One of these on the case side panel I'd image would give some nice whole-of-system cooling.

Put them too close together and the air will just go around the heat sink instead of through it. The whole point of using fins is to expose a hot surface to a steady stream of air.
