Back plates

Are back plates required when water cooling your GPU's or CPU?
The backplate that comes with most CPU waterblocks is required yes.
With a GPU it is not required nor does it come with one usually. But it does help with to keep the card rigid and such. Also really improves the overall look.

I think you may be confusing mounting plates with backplates.

It depends on where the threads are for the screws really.

Yeah, I'm planning on the EK-Supreme HF, I am assuming that it comes with it.

From the EK web shop:

EK-Supreme HF water block
-4 different jet plates + 1 blank
-Universal supported mounting plates:
oIntel: 775, 1366, 115X, xeon
oAMD: AM2, AM2+, AM3, 939, 754, opteron
-Universal CPU retention backplate
-Thermal grease Arctic Cooling MX-2 (2g)

So yes the mounting backplate is included for the above CPU sockets.

Backplates for the GPU are only for bling and serve no purpose other than to look butiful.

This exactly. The required mounting materials for the cpu will come with the block.

GPU (WC) backplates are bling only...if anything, they add more weight for your expansion slot bracket to carry.

Still...look purdy.
GPU backplates barely weigh anything, well the EK ones I have don't weigh much. It also makes the PCB feel a bit more sturdy IMO compared to when it's completely naked or just has a WB on it.
some gpu require a back plate to help cool the ram chips on the other side of the pcb, ie hd5870 2 gb and 5970 2gb, so they do serve a purpose for SOME gpu applications. But for most, as many have mentioned, its just bling and may lend to some rigidity and avoid "bowing" the pcb w/ the strain of the block weight.
