What games are you playing? Don't most games lock the mouse to the current screen and not allow you to go into the second one? (Unless you alt+tab or alt+esc)

FPS and WoW mostly, right now I play WoW in Windowed Fullscreen so I can ALT-TAB to get to iTunes, would switch to Fullscreen if I can get this going.
But is there a way to set a macro to switch the mouse to the other screen?

EDIT: I'd also like to lock my screen so I can't accidentally drag windows into that screen.

I'm not sure honestly. I would assume the best way to scroll through songs would be media keys. If your keyboard has built in macros(assuming it does) it should have media keys. What kind of keyboard do you have?

Unfortunately, there is no way to get your games in full screen with iTunes available if all the displays are hooked to the same GPU. If you want to play full screen while having access to iTunes you will need a second GPU to drive the extra display. This is because the GPU will power off any extra displays once the master display goes full screen.

As for mouse management, you could try using programs like Ultramon and DisplayFusion for multi-display hot keys and task bars. DisplayFusion is my personal favorite but the Pro version, full featured, costs money while Ultramon is free.

Just for my own knowledge, does this work if the game you're playing is utilizing SLI?

Does what work? If you are running in SLI, but still only using 1 or 2 displays, it is still going to exhibit the same behavior since all the displays, unless your are running surround, will be hooked up to the same GPU.
