Oooooh you bet! When I made the move to crossfire I had way too much fun, haha. Feeling the excitement - makes me want to upgrade again!.. Hopefully Bulldozer is worthy, or else I'll follow you on to Sandy Bridge real soon here

No, there is no realistic difference, they usually just suggest the slots that are "read" first if I'm not mistaken. There wouldn't be a performance difference if you were to put another 8GB in the other 2 slots (i.e. slots A/B2 wouldn't be faster than A/B1).
 
As DeltaUpsilon has quoted from the User Manual, the two black or the two blue slots can be used. But Asus suggests using the blue slots (A2 and B2) for better compatibility. It makes no mention of performance reason. But what this compatibility is referred to? Perhaps between the memory controller and the DIMMs? Asus does not elaborate.

Asus forum moderator Ulrich gave the following response to a similar question on P8P67 Deluxe:-

The first black slot is A1 .. that is the closest to the processor.. then comes A2 the Blue one.. Then B1.. a black slot... then B2 a blue slot. The memory controller apparently looks at A2 first.. so Asus recommends starting with a module in A2.. the second slot.. a blue one. Then a second module in B2... the other blue slot.

So if you have your memory in the two blue slots, you have it installed correctly for dual channel operation.
